چکیده

Objective: The objective of the study was to evaluate the dose dependent effects of atorvastatin on rat kidneys. Methods: The selected doses of atorvastatin were administered orally for 17 days to rats. The statin induced nephrotoxicity was accessed by carrying out renal function tests and is by renal histopathology analysis. Measurements of antioxidant enzymes were also carried out to check whether atorvastatin at the selected dose level interfere with any of the antioxidant system. Atorvastatin at three doses 5mg/kg, 10mg/kg and 20mg/kg was tested. Results: Higher doses of atorvastatin exhibits considerable degree of renal toxicity in rats, while the low doses do not interfere with renal functions in rats. Atorvastatin at all the tested dose levels not interfere with cellular anti-oxidant mechanisms. Conclusions: Atorvastatin at 20mg/kg exhibit significant toxicity on rat kidneys. Atorvastatin at the selected dose level does not interfere with any of the antioxidant mechanisms. It indicated that atorvastatin not cause oxidative stress induced renal damage and rhabdomyolysis may be the reason for renal damage induced by atorvastatin at the high dose level. Effect of Simvastatin on Cisplatin-induced Nephrotoxicity in Male Rats

Statins have antioxidant and anti-inflammatory effects that are not directly related to their cholesterol-lowering activity. This study aimed to investigate the effect of simvastatin on the extent of tissue damage in cisplatin-induced nephrotoxicity. Simvastatin was orally given to rats in different doses (1, 2 and 4 mg/kg), 1 h prior to cisplatin injection (5 mg/kg, i.p.). All animals we...

Dose-Dependent Amelioration of Gentamicin-Induced Nephrotoxicity in Adult Swiss Albino Rats by Vitamin B-complex - A Preliminary Study

Purpose: To evaluate the effect of vitamin B-complex on the nephrotoxicity of gentamicin in an established rat model. Methods: Adult Swiss albino rats weighing 170±20g were divided into 4 groups of 4 rats each. Each group was given one of the following: placebo injection (Control), 80mg/kg of gentamicin sulphate alone or with 1.5ml/kg/3ml/kg body weight of vitamin B-complex (intramuscular) cont...

Nephroprotection of lacidipine against gentamycin-induced nephrotoxicity in albino rats.

AIM Gentamycin, a widely-used aminoglycoside antibiotic, is recognized as possessing significant nephrotoxic potential in human beings. Gentamycin-induced nephrotoxicity is suggested to be mediated via reactive oxygen species. The present study investigated the possible antioxidant nephroprotective effect of lacidipine as a calcium-channel blocker in a gentamycin-induced nephrotoxicity model in...

Protective effects of aqueous extract of M. Pruriens Linn. (DC) seed against gentamicin induced oxidative stress and nephrotoxicity in rats

In the present study, we investigated the effects of the aqueous extract of Mucuna pruriens against gentamicin (100 mg kg -1 per day for six days) induced oxidative stress and nephrotoxicity in rats. Administration of gentamicin to rats induced a marked renal failure, characterized by a significant increase in urine volume and serum creatinine and urea levels. There was a significant decr...

Protective Effect of Safranal against Hexachlorobutadiene-Induced Nephrotoxicity in Rat

Background: Hexachlorobutadiene (HCBD) is a potent nephrotoxin in rodents, which can cause degeneration, necrosis and regeneration in renal tubular epithelial cells. It has been shown that safranal, the active ingredient of saffron, has a protective effect against ischemic injuries. The aim of this study was to examine the protective effect of safranal against HCBD-induced nephrotoxicity in rat...
